Laccase has been the focus of considerable interest in recent years, especially for its possible potential use in a variety of industries. Application of laccase is useful in the textile, paper, cellulose and petrochemical industries, where the disposal of waste and its impact on the environment is a considerable problem.

Application

LentiKatsÒ Laccase Production System for large scale production of laccase uses recombinant yeasts immobilized inside the Lentikats Biocatalyst TtL. The Lentikats Biocatalyst TtL is subsequently used by the end user, as a production system during the (A) production of immobilized laccase in Lentikats Biocatalyst TtL or (B) production of free laccase in a liquid medium by a system of repeated batch conversions using a system of three stainless steel reactors.

Fully activated (fully colonized) Lentikats Biocatalyst TtL is used in repeated batch conversions for the production of laccase which is then released into the reaction broth. In each cycle, the production reactor (R1) is filled with production medium and during the production batch laccase is secreted into the reaction medium. The medium containing laccase is then pumped to the deactivation reactor (R2), where the native yeast cells are deactivated/killed. The production reactor (R1) is subsequently fed with fresh production medium. 96% v/v ethanol is added to the deactivating reactor (R2) to a final concentration of 20% and all native yeasts cells are deactivated. Medium containing produced laccase and inactivated free yeast cells is subsequentlydrained out of the reactor R2 and collected in a storage tank (R3). After filling the storage tank (R3) with broth, it can be used as a liquid solution of laccase enzyme directly for further reactions, or purified/concentrated using appropriate downstream processes. At steady state, the system is able to produce laccase activity in range of 28-32 U/L. Medium containing the enzyme laccase can be used directly in certain industries or may be purified and further concentrated for transportation to other end users.

 

Note

The LentiKatsÒ  Laccase Production System uses Lentikats Biocatalyst with immobilized genetically modified (GM) yeast.
